Amanda Kohler, PhD

Dr. Amanda Kohler leads our protein engineering efforts as a consultant with Tyrala Consulting, bringing more than 15 years of experience in protein engineering, recombinant protein production, and antibody discovery to Mitera Biosciences. With early training as an X-ray crystallographer and a strong commitment to applied science, she leverages structure-guided protein design and therapeutic engineering to advance programs from early concept to development-ready biologics.


Across roles of increasing responsibility in biotech and biopharma, Amanda has supported more than a dozen development pipelines in oncology and autoimmune disease, leading early-stage antibody discovery, establishing robust recombinant production systems, and guiding functional and developability assessment to enable data-driven lead selection. Her approach is modality-agnostic and centered on matching therapeutic design to the intended mechanism of action. Her work has spanned development of monoclonal antibodies, bi- and multi-specifics, recombinant proteins, ADCs, radiotherapeutics, and small molecule therapeutics.


Dr. Kohler is an inventor on multiple patents and a co-author of peer-reviewed publications in PNAS, Cell Chemical Biology, JACS, and other journals. Her leadership emphasizes scientific rigor, clear communication, and strong cross-functional collaboration, helping teams make confident decisions in the development of next-generation therapeutics. She holds a Ph.D. in Biological Sciences, Structural Biology and Biochemistry from The Rockefeller University and a B.S. in Biochemistry and Molecular Biology from the University of California, Davis. Prior to consulting, she held scientific roles at Plexxikon and CytomX Therapeutics and served as Associate Director of Protein Engineering at Earli.
